The use of the biological indicator Bacillus stearothermophilus can assure the efficiency of the autoclave
Sterikon (Merck) is used for effectiveness of steam sterilization (autoclave) at 121oC for 15 min.
Pink color - indicates adequate sterilization, whereas growth
Yellow color - shows inadequate sterilization
This bioindicator consists of an ampule containing broth, sugar, pH indicator and spores of Geobacillus steraothermophilus.
The thermal resistant shows that the spores are totally killed after 15 min when heated 121oC in a compressed steam (1 bar). At lower temperatures or lower exposure times the spores can survive.
How to use:
- One ampule is placed into the autoclave.
- Incubate 2 ampules (the autoclave ampule, test) and one ampule (no autoclaved, control) at 60oC for 24-48h.
- No grow of G. stearothermophillus (pink color) indicates adequate sterilization, whereas growth (yellow color) shows inadequate sterilization. The control will show yellow color.
- This procedure should be repeated 3 times a year.
